IHC typically involves simultaneous detection of multiple tissue antigens. To avoid cross-reactivity of secondary antibodies with primary antibodies, researchers typically use antibodies from different species. One problem is that antibodies raised in different animal species to a particular antigen might not be available. You CAN use primary antibodies raised in the same animal species if the antibodies contain different labels, for example, with biotin and DIG, and the detection reagents provide different colors.
